I did the same trip a couple of years ago. I actually left my car at the
Eugene bus station and took Amtrak to Astoria, though I have heard
you can leave your car at the tourist info building in Astoria. I would
give them a call to confirm.
Getting out of Brookings was a pain, and I had to spend a couple of
extra days there because the bus only ran during the week. I had to
transfer in Coos Bay. It was a hassel.
I would suggest reserving a Uhaul. It is a lot cheaper, since rental car
companies charge through the nose for one-way, and they often don't
want you loading a bike in their cars. There was not a u-haul facility in
Brookings. I think the closest was Cresent City, which is not that much
further down the Coast. Try and make a reservation now, though. I
tried to get one with a day's notice and they were booked.
It is a wonderful trip. If you are giving yourselves time to tour around
some of the towns, make sure to check out the aquarium in Newport.
Also in Newport is the Rogue brewery, where you can have lunch
overlooking the bay. One of my favorite spots.

cheers-
melissa

my wife and I rode from Seattle to Crescent City, CA.
We rented a car in Crescent City and drove back. The one way rental was the most convient and cost effective return method for us.
